Treatment Response in GBM Cells Under Normoxic and Hypoxic Conditions

Abstract AIMS Glioblastomas (GBMs) are characterised by highly hypoxic regions and the ability to invade into healthy brain tissue promoting tumour dissemination recurrence; development of novel anti-migratory inhibitors target cell invasion recurrence must consider varying treatment responses in this background. As proof principle we investigated activity two, previously characterised, small molecule under normoxic conditions. We hypothesised that drug is dependent on hypoxia levels. METHOD Glioma line U251 was exposed BIO-indirubin Y-27632 alone or combination assessed 2D scratch assays (21% O2) (0.1% Live imaging performed analysis carried out using ImageJ. RESULTS Treatment with BIO, had a statistically significant effect inhibition migration normoxia (p=0.01); BIO inhibited promoted normoxia, whereas maintained hypoxia, switched towards potentiated. CONCLUSION An induced conditions suggesting adoption amoeboid allowed targeting migratory pathways control RhoA ROCK.
